Abstract:

The effects of chronic stress and glucocorticoids receptor antagonist (RU486) on expression of melanocortin 4 receptor (MC4R) mRNA in arcuate nucleus (ARC) of male rats were evaluated. In this study, adult male Sprague Dawley rats were placed into four groups (n=6/group) stress, RU486, stress/RU486, and control groups. In stress group, the rats were restrained, 1 h/day, for 12 days. In RU486 group, the rats were injected RU486 for 12 days. In stress/RU486 group, the rats were injected RU486 1 h before the stress process for 12 days. Relative expression of MC4R mRNA was determined using real-time PCR. Relative expression of MC4R mRNA in the stress group was higher than that of the control rats (P0.05). Chronic restraint stress causes increase in mRNA expression of MC4R in ARC and blockade of glucocorticoid receptors has no effect on this up-regulation.

P-21: RF-Amide Related Peptide mRNA Expression in Male Rat Dorsomedial Hypothalamic Nucleus during Chronic Stress

Background Chronic stress induces male reproductive dysfunction; however, the involved mechanisms are not clear. It has been suggested that the gonadotrophin-inhibitory hormone, known as RF-amide related peptide (RFRP) in mammalian species, may mediate this inhibitory effect of stress. The objective of the present study was to compare the effect of chronic stress and glucocorticoids receptor an...

P-5: Effect of Chronic Stress on Kiss-1 mRNA Expression in Male Rat Arcuate Hypothalamic Nucleus

Background Kisspeptin is a potent stimulator of the hypothalamic- pituitary-gonadal axis and may play a critical role in modulating of stress-induced suppression of reproduction. The aim of the present study was to investigate the effect of glucocorticoid receptor antagonist (RU486) and stress on expression of Kiss-1 mRNA in arcuate nucleus (ARC) of rat hypothalamus. MaterialsAndMethods Twenty ...

Hypothalamic Expression of Melanocortin-4 Receptor and Agouti-related Peptide mRNAs During the Estrous Cycle of Rats

Melanocortin- 4 receptor (MC4R) and agouti- related peptide (AgRP) are involved in energy homeostasis in rats. According to MC4R and AgRP effects on luteinizing hormone (LH) secretion, they may influence the estrous cycle of rats. Therefore, the aim of this study was to investigate the expression of MC4R and AgRP mRNAs at different stages of estrous cycle in the rat’s hypothalamus. The estrous ...

Melanocortin 4 receptor distribution in the human hypothalamus.

OBJECTIVE The melanocortin 4 receptor (MC4R) is an essential regulator of energy homeostasis and metabolism, and MC4R mutations represent the most prevalent monogenetic cause of obesity in humans known to date. Hypothalamic MC4Rs in rodents are well characterized in neuroanatomical and functional terms, but their expression pattern in the human hypothalamus is unknown. DESIGN AND METHODS To d...
